Here, we show that lipid droplets are associated with proteins involved in fusion processes in the cell: NSF (N-ethylmaleimide-sensitive-factor), alpha-SNAP (soluble NSF attachment protein) and the SNAREs (SNAP receptors), SNAP23 (synaptosomal-associated protein of 23 kDa), syntaxin-5 and VAMP4 (vesicle-associated membrane protein 4).
In two papers in this issue of PNAS, Rothman and colleagues (1, 2) demonstrate that lipo-somes reconstituted with only v- and t-SNAREs can fuse and mix luminal contents with reasonably physiological kinetics. These results further implicate SNAREs as the core fusion machinery for intracellular vesicle trafficking.
Protein Determinants of SNARE-Mediated Lipid Mixing
2010年7月21日 · In this article, we intend to comprehensively characterize the determinants of SNARE-mediated lipid mixing in the context of the FRET-based liposome fusion assay using active proteins. We carry out systematic titrations with proteoliposomes prepared at various protein densities and using different reconstitution strategies.
